• Thymidine Kinase Activation: Aciclovir has a strong affinity for an enzyme known as thymidine kinase (TK) which is produced by the Herpes simplex and varicella-zoster viruses. When this medicine enters infected cells, it gets activated by this enzyme.
• Inhibition of Viral DNA Polymerase: Once activated, aciclovir competes with deoxyguanosine triphosphate (dGTP), a building block of DNA, for incorporation into the growing viral DNA chain. When incorporated into the viral DNA chain, aciclovir results in premature termination of the chain and blocks further replication of the virus.
