• Antiretrovirals such as rilpivirine, atazanavir, and nelfinavir: Vonoprazan reduces stomach acidity, potentially altering the absorption of these antiretroviral medications, which could affect their effectiveness and safety.
• CYP3A Substrates such as verapamil or cyclosporine: Vonoprazan weakly inhibits CYP3A enzymes, which may increase the concentration of CYP3A4 substrates in the body, potentially leading to adverse reactions.
• CYP2C19 Substrates such as clopidogrel, citalopram, and cilostazol: Vonoprazan inhibits CYP2C19 enzymes, which can alter the metabolism of these medicines, affecting their efficacy. 
• Strong or Moderate CYP3A4 Inducers such as phenytoin: Strong or moderate CYP3A inducers reduce Vonoprazan exposure, potentially reducing its effectiveness in treating conditions for which it is prescribed. 
• Other medications such as iron salts: Vonoprazan may reduce the absorption of iron, impacting its therapeutic benefits.

• Gastric Malignancy: Vonoprazan may mask symptoms of gastric malignancy (stomach cancer), delaying diagnosis and treatment.
• Acute Tubulointerstitial Nephritis: Vonoprazan may increase the risk of acute tubulointerstitial nephritis, a kidney disorder, especially with prolonged use.
• Clostridioides difficile-Associated Diarrhea (CDAD): Use of Vonoprazan may increase the risk of Clostridioides difficile-associated diarrhoea, a potentially severe infection of the colon.
• Bone Fracture, including Osteoporosis-related Fracture: Long-term use of Vonoprazan may increase the risk of bone fractures, including those related to osteoporosis, due to potential effects on calcium absorption and bone density.
• Vitamin B12 (Cobalamin) Deficiency: Vonoprazan can reduce the absorption of vitamin B12, leading to deficiency over time, especially in long-term users.
• Hypomagnesemia and Mineral Metabolism: Prolonged use of Vonoprazan may lead to hypomagnesemia (low magnesium levels) and affect mineral metabolism, which can have various health implications.
• Fundic Gland Polyps: Vonoprazan use has been associated with an increased risk of fundic gland polyps, which are benign growths in the stomach.
