Tacrenolime 0.25 Capsule 10 is used for the following:

• Prevention of organ rejection in transplant patients

• Management of atopic dermatitis (eczema)

• Treatment of rheumatoid arthritis

• Management of nephrotic syndrome

• Treatment of psoriasis

• Management of uveitis (eye inflammation)

• Prevention of graft-versus-host disease

• Management of severe autoimmune conditions

• Treatment of resistant inflammatory conditions

• Prevention of transplant failure

• Management of immune system disorders

Follow these instructions while taking Tacrenolime 0.25 Capsule 10:

• Take the capsule on an empty stomach, at least 1 hour before or 2 hours after meals.

• Swallow the capsule whole with a full glass of water.

• Do not crush, chew, or open the capsule.

• Take the medicine regularly at the same time each day.

• Do not stop taking this medicine without consulting your doctor.

• Store in a cool, dry place away from direct sunlight.

Tacrenolime 0.25 Capsule 10 is taken orally.

Tacrenolime 0.25 Capsule 10 contains tacrolimus (0.25 mg), which works on the body as follows:

• FKBP-12 binding: Tacrolimus binds to an intracellular protein called FKBP-12 (FK506 binding protein), forming a complex that is essential for its immunosuppressive action.

• Calcineurin inhibition: This medicine blocks an enzyme called calcineurin phosphatase, preventing the activation of T-cells, which are white blood cells involved in immune responses.

• Prevention of cytokine production: By inhibiting calcineurin, tacrolimus blocks the production of important immune system messengers called interleukins (such as IL-2, IL-3, IL-4, IL-5) and other cytokines that activate and help T-cells multiply.

• Anti-inflammatory action: In skin conditions like atopic dermatitis, tacrolimus reduces inflammation similarly to steroids but without causing skin thinning, making it suitable for sensitive areas.

• Store Tacrenolime 0.25 Capsule 10 at room temperature (below 30°C).

• Keep the medicine in its original packaging until use.

• If you've had an organ transplant, maintain good hygiene practices and avoid crowded places during the first few months to reduce your risk of infections, as tacrolimus suppresses your immune system.

• Avoid eating grapefruit or drinking grapefruit juice while taking Tacrenolime 0.25 Capsule 10, as this can increase the amount of tacrolimus in your blood to potentially harmful levels.
