Graforin 50 Capsule helps calm an overactive immune system in serious medical conditions. Uses of Graforin 50 Capsule are:

• Prevention of organ rejection: This medicine stops the body from attacking and rejecting a newly transplanted kidney, liver, heart or other transplanted organ.
• Bone marrow transplant complications: It prevents rejection after bone marrow or stem cell transplantation and manages graft-versus-host disease (a condition where transplanted cells attack the recipient's body).
• Severe rheumatoid arthritis: This medicine manages severe joint inflammation and damage when methotrexate (another medicine) has not worked well enough.
• Severe plaque psoriasis: It manages severe skin plaques (thick, scaly patches) when other treatments have failed or cannot be used.
• Severe atopic dermatitis: This medicine manages severe eczema (a skin condition causing red, itchy rashes) when usual treatments are not effective.
• Sight-threatening eye inflammation: It manages endogenous uveitis, a serious inflammation inside the eye that can damage vision.

Graforin 50 Capsule contains cyclosporine (50 mg), which works in the body as follows:

• Binds to cyclophilin: Cyclosporin enters immune cells and attaches to a receptor protein called cyclophilin, forming a complex inside the cell.
• Blocks calcineurin enzyme: The cyclosporin-cyclophilin complex then inhibits an enzyme called calcineurin, which normally activates immune responses by switching on certain genes.
• Prevents activation of T-cells: By blocking calcineurin, cyclosporin stops a transcription factor (a protein that controls gene activity) called NFAT from entering the cell nucleus and turning on genes for inflammatory signalling molecules.
• Suppresses interleukin-2 production: This action reduces the production of interleukin-2, a key chemical messenger that drives T-lymphocytes (a type of white blood cell) to multiply and attack transplanted organs or the body's own tissues.

• Store below 30° C, away from heat and moisture.
• Keep the capsules in their original container or blister strip until you take them.
• Do not store in damp places like bathrooms.
• Keep out of the sight and reach of children.
• Do not use after the expiry date printed on the pack.

• Maintain good oral hygiene: Brush your teeth regularly with a soft toothbrush and maintain good dental hygiene to help limit gum overgrowth and swelling during treatment.
• Choose a balanced diet: Include a variety of fruits, vegetables, whole grains and other nutritious foods as part of your regular meals to support general wellbeing during long-term treatment.
