Esomeprazole + Levosulpiride is a combination medicine used to treat gastro-oesophageal reflux disease (GERD) that does not respond well to a single acid-reducing medicine.

This combination is commonly used for:

• Gastro-oesophageal reflux disease (GERD): Relieves heartburn, acid regurgitation, and chest discomfort caused by stomach acid flowing back into the food pipe.
• Poorly controlled acid reflux: Used when symptoms persist despite treatment with a proton pump inhibitor (PPI) alone.
• Associated digestive discomfort: Helps ease bloating, nausea, and a feeling of fullness linked to slow stomach emptying.

How Esomeprazole + Levosulpiride works

Esomeprazole + Levosulpiride works by reducing stomach acid production and improving the movement of food through the digestive tract.

Esomeprazole is a proton pump inhibitor. It blocks an enzyme in the stomach lining called the proton pump, which is responsible for producing acid. This reduces the amount of acid in the stomach, easing symptoms such as heartburn and acid regurgitation.

Levosulpiride works differently. It blocks dopamine (D2) receptors in the digestive tract, increasing the release of acetylcholine. This improves stomach emptying, strengthens the lower oesophageal sphincter, and reduces the backward flow of stomach acid. It also has a mild effect on nausea and vomiting.

Together, these two medicines reduce acid production while improving digestive movement, providing more complete relief from reflux symptoms.

Contraindications

  • The concurrent use of medications, including painkillers (aspirin, naproxen), anti-HIV medications (atazanavir), blood-thinning medications (clopidogrel, warfarin, cilostazol), anti-fungal medications (ketoconazole, itraconazole, voriconazole), anti-cancer medications (erlotinib, methotrexate), anti-depressants (citalopram, imipramine, clomipramine, duloxetine), anti-anxiety medications (diazepam), anti-convulsants (phenytoin), gastroprokinetic agents (cisapride), medication for heart problems (digoxin), immunosuppressants (tacrolimus), and anti-tuberculosis medications (rifampicin), alongside Esomeprazole + Levosulpiride is not recommended. Prior consultation with a doctor is essential before considering the use of any prescription or over-the-counter medications to ensure the safety and efficacy of the combined treatment plan.

Overdose and Missed Dose

  • Taking more than the prescribed dose may cause confusion, drowsiness, blurred vision, a fast heartbeat, nausea, sweating, flushing, headache, and dry mouth. Inform your doctor if you notice these symptoms and follow their advice.
  • If you miss a dose, take it as soon as you remember. If it is nearly time for your next dose, skip the missed dose and continue with your regular schedule. Do not take a double dose to make up for the one you missed.

  • This combination may affect your ability to drive or operate heavy machinery. Avoid driving if you feel dizzy after taking this combination.

Use in Pregnancy and Lactation

  • Esomeprazole + Levosulpiride is not recommended during pregnancy and while breastfeeding.
