  • Domperidone + Rabeprazole is used in the treatment of Gastroesophageal Reflux Disease (GERD) when patients are not responding to Rabeprazole alone.

How Domperidone + Rabeprazole works

  • Domperidone, a dopamine antagonist with antiemetic properties, exhibits limited permeability through the blood-brain barrier. While extrapyramidal side effects are exceedingly rare in adult users, domperidone stimulates prolactin release from the pituitary. Its antiemetic efficacy may result from a blend of peripheral (gastrokinetic) actions and antagonism of dopamine receptors in the chemoreceptor trigger zone, situated beyond the blood-brain barrier in the area postrema.
  • Rabeprazole is classified as an antisecretory agent, specifically a substituted benzimidazole proton-pump inhibitor, devoid of anticholinergic or histamine H2-receptor antagonist attributes. It suppresses gastric acid release by inhibiting the gastric H+ and K+ ATPase located at the secretory surface of the gastric parietal cell. Given that this enzyme functions as the acid (proton) pump within the parietal cell, rabeprazole is designated as a gastric proton pump inhibitor. Its mechanism involves blocking the ultimate stage of gastric acid secretion.

Overdose and Missed Dose

  • Overdose of this combination can cause agitation, altered consciousness, convulsions, disorientation, somnolence, and extrapyramidal reactions. Inform your doctor regarding these effects and follow the advice accordingly.
