Uses of Teicoplanin

Teicoplanin is a glycopeptide antibiotic used to treat serious bacterial infections, particularly those caused by gram-positive bacteria, including resistant strains such as MRSA (methicillin-resistant Staphylococcus aureus).

Teicoplanin is commonly used for:

• Skin and soft tissue infections: Treats severe bacterial infections of the skin, including infected wounds and cellulitis caused by gram-positive bacteria.
• Bone and joint infections (osteomyelitis and septic arthritis): Used for bacterial infections of the bones and joints requiring intravenous antibiotic treatment.
Endocarditis: Treats bacterial infection of the inner lining and valves of the heart.
Pneumonia: Manages hospital-acquired pneumonia and ventilator-associated pneumonia caused by susceptible gram-positive bacteria.
Urinary tract infections (complicated): Treats serious UTIs involving susceptible gram-positive bacteria.
Peritonitis and intra-abdominal infections: Used for serious abdominal infections, including those associated with peritoneal dialysis.
Septicaemia (bloodstream infection): Administered for life-threatening bloodstream infections caused by gram-positive organisms.
MRSA infections: Effective against MRSA, making it an important treatment option for susceptible infections caused by this organism.

How Teicoplanin works

Teicoplanin works by targeting and disrupting the formation of the bacterial cell wall, which is essential for bacterial survival.

It belongs to the glycopeptide class of antibiotics. Bacteria build their protective outer wall by linking together building blocks called peptidoglycan precursors. Teicoplanin works by binding tightly to specific components of these precursors the D-alanyl-D-alanine terminal sequences before they can be incorporated into the growing cell wall.

By physically blocking these building blocks, Teicoplanin prevents two essential processes in cell wall assembly:

  • Transglycosylation: The linking of peptidoglycan chains that form the backbone of the cell wall.
  • Transpeptidation: The cross-linking of these chains, which gives the cell wall its strength and rigidity.

Without a complete, properly assembled cell wall, bacteria cannot maintain their structural integrity, swell under internal pressure, and ultimately die.

Teicoplanin is effective against gram-positive bacteria, including Staphylococcus aureus (including MRSA), Streptococcus species, and Enterococcus species. It is not effective against gram-negative bacteria, which have a different outer membrane structure.
