Amorolfine

Antifungal

Uses of Amorolfine

Amorolfine is an antifungal medicine used to treat fungal infections of the nails (onychomycosis) and certain fungal skin infections.

Amorolfine is commonly used for:

  • Nail fungal infections (onychomycosis): Treats fungal infections of the fingernails and toenails, including discolouration, thickening, brittleness, and crumbling of the nails caused by dermatophytes and other fungi.
  • Ringworm (tinea corporis): Clears circular, scaly rashes on the skin caused by fungal infection.
  • Athlete's foot (tinea pedis): Treats fungal infection between the toes and on the soles of the feet, causing itching, redness, and peeling.
  • Jock itch (tinea cruris): Manages fungal infection in the groin area causing itching and a red, circular rash.
  • Pityriasis versicolor: Treats a fungal condition that causes patchy discolouration of the skin.
  • Cutaneous candidiasis: Treats fungal skin infections caused by Candida species.

Side effects of Amorolfine

  • Redness
  • Itching
  • Burning sensation

How Amorolfine works

  • Amorolfine works by targeting the cell membrane of fungi, disrupting essential processes needed for fungal survival and growth.
  • Fungi depend on a substance called ergosterol to maintain the structure and function of their cell membranes. Ergosterol is unique to fungi and is not found in human cells, which makes it an effective target for antifungal treatment.
  • Amorolfine blocks two key enzymes involved in the production of ergosterol: delta-14 reductase and delta-7-8 isomerase. When these enzymes are blocked, ergosterol cannot be produced in normal amounts. Instead, abnormal sterols accumulate in the fungal cell membrane, leading to changes in its permeability and structure.
  • This disruption interferes with critical processes inside the fungal cell, ultimately preventing the fungus from growing and reproducing. Over time, the fungi die off, allowing healthy nail or skin tissue to grow back.
  • Amorolfine has both fungistatic (growth-stopping) and fungicidal (fungus-killing) properties, making it effective against a wide range of fungi including dermatophytes, yeasts, and moulds.

Expert Advice

Dosage Forms Available

  • Amorolfine is used topically (Cream, lacquer).

Age and Dose Restrictions

  • It can be used by adults and children above 12 years of age.

Contraindications

  • It is contraindicated in patients with a history of hypersensitivity to amorolfine.

Other Medications To Avoid With This Medicine

  • It could interact with medications used for respiratory conditions (such as budesonide, formoterol), drugs for HIV/AIDS treatment (like ritonavir, cobicistat), and corticosteroids.

Overdose and Missed Dose

  • If you miss an application of Amorolfine, apply it as soon as you remember. If it is nearly time for the next scheduled application, skip the missed one and continue with your regular schedule. Do not apply a double amount to compensate. For nail infections, consistent and regular application is essential throughout the full prescribed course to achieve effective results.
  • Amorolfine is applied to the nails or skin and is not taken orally, so a systemic overdose is unlikely with normal use. If Amorolfine nail lacquer or cream is accidentally swallowed, contact your doctor or seek medical advice immediately. Do not induce vomiting unless specifically instructed to do so by a medical professional.

Management of Side Effects

  • The side effects due to amorolfine resolve on its discontinuation and do not require medical attention. If you experience severe or persistent side effects from this cream, consult your doctor.

  • Avoid scratching the affected skin area to prevent fungus from spreading to other body parts.
  • Steer clear of sharing personal items like towels, combs, bedsheets, shoes, or socks with others.
